Creation: SC 527 appeared in 1939 as new primary routing, running from US 521 (now SC 377) Kingstree southeast for 10 miles. This is the only SC 527.
Adjustments: In 1940, SC 527 was extended east as new primary routing to end at SC 511 (now SC 41) Warsaw. Also, SC 527 was extended west, replacing SC 175 through Sardinia to end at SC 54 (now US 378).

In 1942, SC 527 was extended northwest as new routing to th Lee-Sumter line, then replaced SC 347 out to Elliott and replaced SC 341 ALT to Wisacky.

In 1947, SC 527 was reduced to a two-piece route: SC 341 to US 76 and from SC 54 to Kingstree. The decommissioned portions became S-31-19; S-42-30; S-45-17 and S-45-21.
In 1949, SC 527 was restored in its entirety from Wisacky to Warsaw.

In 1967 or 1968, SC 527 was placed on new roadway around the south of Kingstree. The old route became SC 527 CONN; a bit of SC 377; SC 261 Main St

SC 527 was multilaned on its US 52 duplex by 1963. It is also multilane from SC 377 to the SCL of Kingstree.
Comment: SC 527 follows the Black River for much of its length.
