Creation: Appeared in late 1926 as an original US Highway as an addition to SC 41 from US 17-76 Florence to end at US 1 (now S-13-148 Cash Rd) Cheraw.
Adjustments: In 1928, SC 41 was removed from US 601.
In 1931, US 601 was removed from S-13-494 Macedonia Church Rd and Juniper road south of Cheraw and placed on modern US 52 to modern US 1.
In 1932 (bridge date), US 601 was removed from Creek Crossing Dr and Pine Log Cir (no longer crosses Black Creek) belwo Dovesville.
In 1933, US 601 was extended replacing SC 96 into North Carolina.
In 1935, US 601 was renumbered as US 52, removing US 601 from South Carolina altogether.

In 1951, US 601 was rerouted in North Carolina and then extended back into South Carolina, replacing SC 151 to Pageland; following SC 9 west a little; replacing SC 902 and SC 265 to Kershaw; added to US 521 and US 1 to Camden and Lugoff; replacing SC 26 to Orangeburg; added to US 301 to Bamberg; replaced SC 36 to US 321 near Pineland; followed US 321 to end at US 17 Hardeeville.

In 1954, US 601 was rerouted in Orangeburg to its current routing by staying on Magnolia a little further. Russell St became an extended SC 33.

In 1965 (bridge date), US 601 was removed from Wintertide Dr and S-13-823 in the Lynches River area. The 1922 bridge over the river is still accessible.

About 1974, US 601 was extended south to I-95 as a multiplex with US 17.
In 1976, US 601 south was truncated to its current US 321 endpoint leaving behind US 321 and US 17.

Sometime after 1991, US 601 appears to have been placed on US 521's bypass of Kershaw, then used S-29-62 Hilton St to get back to US 601's orginal route. Main St became a posted US 601 Business. Note that SCDOT maps do not show this despite it being heavily posted this way.
Improvements: Paved only from Florence to Darlington in 1928, then to Society Hill in 1932; fully paved to Cheraw in 1934. When US 601 returned to S.C. it was fully paved everywhere.
The first multilane section of US 601 appeared 1962, from SC 4 to SC 33.
The next sections to be upgraded between 1968-69 was from Camden to I-20
In 1970, US 601 was multilaned SC 4 to SC 70.
In 1973, US 601 was 4-laned from Orangeburg to I-26 and from SC 70 to the US 301 SB split.
In 1974, US 601 was 4-laned from Camden to SC 97.
In 2007 or 2008, US 601 was multilaned from I-26 to the Calhoun County line.
In 2013, US 601 was multilaned from Orangeburg-Calhoun line to US 176.
Comment: US 601 is little changed from its reintroduction to the Palmetto State.
There is a posted US 601 TRUCK for Camden that uses S-28-130 Springdale Dr/Boykin Rd and serves as a natural bypass for Camden.
